Summary

B7-33 is a single-chain analogue derived from the B-chain of human relaxin-2 (H2 relaxin). It is studied as an experimental, functionally selective ligand of relaxin family peptide receptor 1 (RXFP1); the published evidence is centered on cellular and animal models.

In the reported work, B7-33 bound RXFP1 and preferentially activated ERK1/2-related signaling over cAMP in RXFP1-expressing cells. The described mechanism involved RXFP1–angiotensin II type-2 receptor heterodimers and the collagen-degrading enzyme MMP-2, and the peptide prevented or reversed organ fibrosis in preclinical rodent models of heart and lung disease.
